| I. Background | |
| GARP（Glycoprotein A Repetitions Predominant）與 Latent TGF-β1（轉(zhuǎn)化生長(zhǎng)因子β1 的 潛伏形式）是免疫調(diào)節(jié)中密切關(guān)聯(lián)的關(guān)鍵分子，共同參與維持免疫穩(wěn)態(tài)及疾病發(fā)生發(fā)展。 GARP 基因位于人類染色體 9q34.3，編碼一種跨膜蛋白，屬于亮氨酸富集重復(fù)序列（LRR） 家族，其結(jié)構(gòu)包含多個(gè) LRR 結(jié)構(gòu)域，賦予其結(jié)合特定配體的能力。Latent TGF-β1 是 TGF- β1 的前體形式，由 TGF-β1 前體蛋白經(jīng)加工形成復(fù)合物，包含成熟的 TGF-β1、潛伏相關(guān)肽 （LAP）及可能的 GARP 蛋白，處于無(wú)活性狀態(tài)，需激活后才能發(fā)揮生物效應(yīng)。 GARP 的核心功能是作為細(xì)胞表面受體，結(jié)合并呈遞 Latent TGF-β1。在調(diào)節(jié)性 T 細(xì)胞 （Tregs）中，GARP 高表達(dá)，通過(guò)錨定 Latent TGF-β1 于細(xì)胞表面，形成 GARP-Latent TGF-β1 復(fù)合物。當(dāng) Tregs 受到整合素等信號(hào)刺激時(shí)，Latent TGF-β1 可被激活，釋放成熟的 TGF-β1，從而抑制周圍免疫細(xì)胞的活化，維持免疫耐受，防止自身免疫反應(yīng)。 |
